what is a good vacuole
Vacuolar morphologies in hyphae that are not in contact with nutrients. Conidia of TPVEV4 were inoculated on the side of approximately 1-mm-thick
CD containing 2% agar, and hyphae were allowed to grow in lateral directions for 48 h (a to e, g, h) or 72 h (f, i) by preventing the
upward-directed growth with a cover glass. Vacuolar and/or cell wall-bound invertase isoforms are also induced by de novo injuries to beet tissue
(Milling et al., J. Bot., 44 (1993), 1687?1694).

Cellulose provides structural support while remaining flexible; flower petals, for instance, are flexible but will snap if folded too far.
Cells carrying a vat2 null allele were viable; however, these cells were severely defective for growth in medium buffered at neutral pH. Vacuoles
isolated from cells bearing the vat2 null allele were completely devoid of vacuolar ATPase activity. Cells should be transferred every three
days. It seems that with 95 x 15 mm plates, you can keep a culture going by transferring once a week.
Cells are always wearing out. They are then replaced by new ones. Cell lysates were sonicated, centrifuged at 10,000 xg and analyzed by
Western blot analysis with anti- myc antibodies. There was no significant difference in the expression.

GFPs degradation was much higher in Nicotiana spp. The version of GFP used appeared not to be a good vacuolar marker for Nicotiana
differentiated tissues, although it can efficiently label vacuoles in protoplasts or calli. GFP-Chi is mainly in PSVs, but often labels LVs (g).
GFP-Chi is limited to the ER of expanding root hairs (h) but sometimes labels their vacuole once fully expanded (i).lysosomes, lysosomal
Lysosomes maintain an internal acidic environment through the use of a hydrogen ion pump in the lysosomal membrane that drives ions from the
cytoplasm into the lumenal space of the organelles. The high internal acidity is necessary for the enzymes contained in lysosomes to exhibit
their optimum activity. Lysosomes are the cells' garbage disposal system. They degrade the products of injestion, such as the bacterium that has
been taken in by phagocytosis seen in the above cartoon.

Aphids then allow the internal fluid pressure to force food into its gut through a food tube while the mosquito sucks blood after preventing
blood coagulation by using an anticoagulant saliva. The vampire bat is another good example which laps blood after using its razor-sharp teeth to
cause a wound in its usually sleeping prey. Apoplast targeting is the best approach.
plant,plants
Plant cells contain salts and sugars in a higher concentration than their surroundings so water is drawn in - the cell membrane and the
cytoplasm within the cell act as the semi-permeable membrane. On the fine roots the outer cells have very thin walls and elongated projections or
root hairs which sit between the soil particles. Plant cells have larger vacuoles than animal cells. Discuss what they are and the job they
perform for the cell.

Note the vacuole at the arrow and profiles of similar vacuoles at the arrowheads. An example of a curved vacuole (arrow) positioned at the end
of a basal body (BB). Note that the sulphur is located in a different organelle. The elements are retained in situ by an anhydrous freeze
substitution technique developed by our group.
